Effects of Methoxyflurane Anesthesia on Ultrastructure of Rat Lung

Abstract
The author studied the effects of methoxyflurane (2, 2-dichloro-1, l-difluoro-ethyl methyl ether} on the pulmonary ultrastructure experimentally. Nine albino rats, weighing 200250gm, were anesthetized with methoxyflurane for 60 minutes. And then 3 each animals were sacrificed immediatE~ly, 4 hours and 12 hours after anesthesia, respectively. Another 3 rats were alloted for control. bungs were extirpated and examined using both. light acid electron microscopes.
The results were as follows:
1) Light microscopically, lungs showed mild degree of congestion, interstitial edema, thickening of the alveolar septa, and increased alveolar cells. No increased bronchiolar secretion noted.
2) EIectron microscopically, the osmiophilic inclusion bodies of the type II alveolar cells showed numerical increase, pleomorphism, decrease in density, vacuolization, and extrusion of. the osmiophilic inclusion bodies from the type II alveolar cells, immediately after anesthesia onwards. Type II alveolar cells were increased in number, 4 to 12 hours after anesthesia. Alveolar capillary endothelia:! cells showed increased pinocytotic vesicles, dilatatior.~ of the rER and vacuolization. These alveolar capillary changes were seen to disappear gradually 4 to 12 hours after anesthesia. No significant change was noted in the type 1 alveolar cells. On the other hand, alveolar macrophages were increased and showed autophagic vacuoles in the cytoplasms.
3) In conclusion, the author has found that methoxyflurane anesthesia brings about a mild degenerative and/cr adaptive changes in the osmiophilic inclusion bodies of type II alveolar cells. This methoxyfhirane anesthesia also induces a mild degenerative changes of the alveolar capillary endothelial cells which can be restored to normal within 12 hours after anesthesia.
